Venous thromboembolism (VTE) is a common and fatal disease with substantial burden to society. Cumulative evidence suggests a role of the complement system, specifically the lectin pathway (LP), in the pathogenesis of VTE; however, the underlying mechanisms remain elusive. In this thesis, we have investigated the association of mannose-binding lectin (MBL) and mannose-binding lectin -associated serine protease-2 (MASP-2), key components of the lectin pathway, in relation to VTE by using data from the Nord-Trøndelag Health (HUNT) Study and the Tromsø Study respectively.
We found that high plasma levels of MBL and MASP-2 were associated with increased risk of VTE. Genetic analysis revealed a causal role of MBL and MASP-2 on VTE risk. Further functional analysis indicated that MBL-MASP complexes could trigger thrombin generation, a key component of blood clotting process. Our findings suggest that MBL and MASP-2 are potential risk biomarkers for VTE and molecular targets for prevention of VTE.
